TDS-943 is a topical spray-gel formulation containing 4% diclofenac sodium, developed using the proprietary Mika Delivery System (TDS) for the treatment of pain associated with osteoarthritis, particularly of the knee. As a non-steroidal anti-inflammatory drug (NSAID), diclofenac exerts its therapeutic effect by inhibiting the cyclooxygenase enzymes COX-1 and COX-2, thereby blocking the synthesis of pro-inflammatory prostaglandins. The TDS-943 formulation is designed to provide rapid transdermal penetration and localized concentration of the active ingredient in the underlying tissues and synovial fluid while maintaining low systemic absorption. This targeted delivery profile aims to provide efficacy comparable to oral NSAIDs while significantly reducing the risk of systemic adverse events, such as gastrointestinal and cardiovascular complications. TDS-943 has undergone Phase 3 pivotal testing and is currently marketed in several European territories, although its development for the United States market was discontinued following strategic business decisions by the sponsor.
