TDT-067 is a novel topical antifungal formulation developed by Celtic Pharma Development Services for the treatment of onychomycosis (fungal nail infection). The drug consists of terbinafine (1.5%) encapsulated within a proprietary drug delivery system known as Transfersomes. Transfersomes are ultradeformable lipid vesicles composed of a phospholipid bilayer and a membrane-softening agent (Tween 80), which allows them to penetrate the stratum corneum and nail plate to deliver the active ingredient directly to the nail bed and surrounding tissues. Terbinafine acts by inhibiting squalene epoxidase, a key enzyme in the fungal ergosterol biosynthesis pathway, leading to the accumulation of squalene and a deficiency in ergosterol, which results in fungal cell death. TDT-067 is administered as a topical liquid spray twice daily and was designed to provide high local efficacy with negligible systemic exposure compared to oral terbinafine therapy.
