TDZD-8 (4-benzyl-2-methyl-1,2,4-thiadiazolidine-3,5-dione) is a small molecule research compound that acts as a potent and selective non-ATP competitive inhibitor of glycogen synthase kinase 3 beta (GSK-3β), with an IC50 of approximately 2 μM. As a member of the thiadiazolidinone class, it is distinguished by its mechanism of action which does not compete with ATP, potentially offering higher selectivity over other kinases. TDZD-8 has demonstrated significant neuroprotective, anti-inflammatory, and anti-arthritic activities in various preclinical models. It has been investigated for its therapeutic potential in conditions such as Alzheimer's disease, glioblastoma, rheumatoid arthritis, and cerebral ischemia/reperfusion injury. The compound works by modulating downstream signaling pathways, including the inhibition of NF-κB and the activation of the ERK/p90RSK pathway, thereby influencing cell survival, inflammation, and apoptosis.
