TE-1146 is an antibody-drug conjugate (ADC) developed for the treatment of multiple myeloma. It consists of six lenalidomide molecules site-specifically conjugated to a reconfigured anti-CD38 monoclonal antibody (daratumumab) using a proprietary multi-arm linker technology. This design enables precise delivery of cytotoxic lenalidomide directly to CD38-overexpressing tumor cells, enhancing cell-killing effects compared to either lenalidomide or daratumumab alone or their combination. The ADC remains intact in circulation and releases lenalidomide upon internalization by target cells, leading to improved efficacy and potentially reduced systemic toxicity. The drug is being developed by Immunwork, with T-E Meds also involved in its development pipeline[1][2][3][4][5].
