Overview

TE-6168 is a one-polypeptide fusion protein drug candidate developed by Immunwork. It consists of an anti-fibrin single-chain variable fragment (scFv) fused to reteplase, the protease domain of human tissue plasminogen activator (tPA). The scFv component specifically binds to fibrin in blood clots without interference from high concentrations of fibrinogen, while the reteplase moiety dissolves clots by promoting fibrinolysis. This targeted design aims to enhance clot dissolution efficiency in conditions such as thrombosis, ischemic stroke, and myocardial infarction while reducing the risk of internal bleeding compared to existing tPA forms like alteplase and tenecteplase. Preclinical studies in mouse and rat models have shown improved clot-targeting ability, thrombolytic activity, and stroke recovery efficacy with milder side effects than current therapies[2][1].
