TE-8214 is an ultralong-acting synthetic peptide analogue of octreotide, developed by Immunwork. It is designed as a somatostatin receptor agonist with a molecular structure consisting of an octreotide derivative extended by a short peptide linker at the N-terminal and conjugated to two fatty acids. This modification enables prolonged activity compared to standard octreotide formulations. In preclinical models, TE-8214 demonstrated sustained inhibition of serotonin secretion in neuroendocrine tumor xenografts and long-lasting suppression of insulin-like growth factor 1 (IGF-1) and growth hormone in animal studies. The drug is being developed primarily for the treatment of acromegaly and neuroendocrine tumors (NETs), with potential advantages over existing somatostatin analogues due to its extended duration, subcutaneous administration with thin needles, and reduced risk of injection site nodules. Phase 1 clinical trials are ongoing in Australia[1][2][3][7].
