StemSynergy Therapeutics is developing small molecule inhibitors of the TEAD-YAP interaction, a central component of the Hippo signaling pathway. The Hippo pathway is a critical regulator of organ size, tissue homeostasis, and cell proliferation. Dysregulation of this pathway, particularly the hyperactivation of the transcriptional co-activator YAP (Yes-associated protein) and its subsequent binding to TEAD (TEA domain) transcription factors, drives the expression of genes that promote tumor growth, survival, and resistance to conventional therapies. By disrupting the YAP-TEAD protein-protein interaction, these inhibitors aim to suppress oncogenic signaling across a variety of solid tumors. The program is currently in the preclinical stage for pan-cancer applications.
