Tebentafusp is a first-in-class bispecific T-cell receptor (TCR) fusion protein immunotherapy designed to treat unresectable or metastatic uveal melanoma in HLA-A*02:01-positive adults. It consists of an engineered TCR domain that specifically binds to a gp100 peptide presented by HLA-A*02:01 on melanoma cells, fused to an anti-CD3 single-chain variable fragment (scFv) effector domain. This structure enables the drug to physically bridge and activate CD3+ T cells against tumor cells expressing gp100, leading to targeted cytotoxicity and tumor cell death through the release of perforins and granzymes. Tebentafusp is only effective in patients who are positive for the HLA-A*02:01 antigen[1][2][6][7]. Developed by Immunocore, it was shown in clinical trials to improve overall survival compared with standard therapies for metastatic uveal melanoma[6][7].
