Tecaginlimab is an Fc-inert, bispecific IgG1 monoclonal antibody that conditionally co-stimulates the CD40 and 4-1BB (CD137) pathways by simultaneously binding CD40 on antigen-presenting cells and 4-1BB on T cells, thereby enhancing anti-tumor immune responses in a localized, crosslinking-dependent manner.[4][7][13][10] Developed using the DuoBody bispecific platform, the molecule is being investigated primarily for advanced solid tumors, including in combination with radiotherapy and checkpoint inhibitors, aiming to achieve potent immune activation with reduced systemic toxicity compared with conventional agonist antibodies.[7][13][18]
