Overview

Tecastemizole (also known as norastemizole) is a potent and selective H1 receptor antagonist and the major active metabolite of astemizole. It was developed as a next-generation antihistamine for the treatment of allergic conditions such as allergic rhinitis. Tecastemizole exhibits anti-inflammatory activity, including inhibition of eosinophil recruitment in models of allergic inflammation, and may have both H1 receptor-dependent and independent mechanisms for suppressing inflammatory responses. Despite promising results in clinical trials up to phase III, its development was discontinued due to adverse effects observed in animal studies, including phospholipidosis and cardiomyopathies[1][4][7][8].
