Overview

Teclistamab + autologous lymphocyte infusion (ALI) is an investigational combination therapy being evaluated for the treatment of relapsed or refractory multiple myeloma (RRMM). Teclistamab is a humanized bispecific T-cell engager (BiTE) that targets B-cell maturation antigen (BCMA) on myeloma cells and CD3 on T lymphocytes, thereby facilitating T-cell-mediated cytotoxicity. The addition of autologous lymphocyte infusions (ALI) aims to replenish or enhance the T-cell pool to improve the duration of response and efficacy of teclistamab, particularly in patients who have achieved at least a partial response after initial teclistamab monotherapy. This specific combination is being studied in the Phase II TALIM trial (NCT06933277) sponsored by the Gruppo Italiano Malattie EMatologiche dell'Adulto (GIMEMA).
