Teclistamab + PD-1 inhibitor is an experimental drug combination for the treatment of relapsed or refractory multiple myeloma and other cancers. **Teclistamab** is a bispecific monoclonal antibody that engages CD3-positive T cells and B-cell maturation antigen (BCMA) on myeloma cells, thereby redirecting T cells to kill cancer cells. **PD-1 inhibitors** (such as pembrolizumab or nivolumab) are immune checkpoint inhibitors targeting the programmed cell death protein 1 (PD-1) on T cells, blocking its interaction with PD-L1/PD-L2 and restoring T cell anti-tumor activity. The rationale behind this combination is to enhance anti-tumor immunity: teclistamab activates T cells against myeloma cells, while PD-1 inhibitors reduce T cell exhaustion, potentially overcoming resistance and producing synergistic anti-myeloma effects. This combination is currently in early-phase clinical evaluation, with ongoing studies to assess efficacy and safety. The combination is primarily developed for multiple myeloma, particularly in patients with relapsed or refractory disease.[1][5][6]
