Tegacorat (GRT6019) is an investigational, orally bioavailable small molecule glucocorticoid receptor modulator (GRM) developed by Grünenthal. It is designed to act as a selective ligand for the glucocorticoid receptor, aiming to provide broad anti-inflammatory efficacy similar to traditional corticosteroids but with a significantly improved safety profile. By modulating the receptor's activity rather than providing full agonism, tegacorat potentially reduces the systemic side effects typically associated with long-term steroid use, such as bone density loss and metabolic disturbances. The drug has completed Phase I clinical trials, including pharmacokinetic and mass balance studies, and is being advanced into Phase II development for the treatment of Duchenne Muscular Dystrophy (DMD).
