TEGAR T cells are an experimental chimeric antigen receptor (CAR) T-cell therapy designed to treat glypican-3 (GPC3)-positive liver cancers, including hepatocellular carcinoma and hepatoblastoma. These are second-generation CAR-T cells engineered via retroviral transduction to express a CAR derived from the GC33 antibody, which specifically recognizes GPC3. A unique characteristic of the TEGAR platform is the co-expression of the cytokines Interleukin-15 (IL-15) and Interleukin-21 (IL-21). These cytokines are intended to improve the metabolic fitness, expansion, and long-term persistence of the T cells within the immunosuppressive tumor microenvironment of the liver, addressing a common limitation of CAR-T therapies in solid tumors.
