Tegideridine (also known as SHR8554) is a novel, small-molecule G-protein biased μ-opioid receptor (MOR) agonist developed by Jiangsu Hengrui Pharmaceuticals. It is specifically designed for the management of moderate to severe acute postoperative pain. By selectively activating the G-protein signaling pathway while minimizing the recruitment of β-arrestin 2, tegideridine aims to provide potent analgesia with a significantly improved safety profile compared to traditional opioids like morphine. This biased signaling mechanism is intended to reduce the incidence of opioid-related adverse events, such as respiratory depression, nausea, vomiting, and gastrointestinal dysfunction (constipation). The drug received marketing approval from China's National Medical Products Administration (NMPA) in May 2023.
