Tegileridine (SHR8554) is a small molecule μ-opioid receptor (MOR) biased agonist developed by Jiangsu Hengrui Pharmaceuticals for the treatment of postoperative pain. It is designed to selectively activate the G-protein-coupled signaling pathway, which mediates central analgesic effects, while exhibiting minimal recruitment of β-arrestin-2, a pathway linked to adverse events such as respiratory depression and gastrointestinal dysfunction. In January 2024, tegileridine fumarate injection received its first approval in China for the management of moderate to severe pain following abdominal surgery, representing the first domestically developed Class 1 opioid analgesic innovation drug in the country.
