Overview

Tegituridine (HS-10388) is a novel small-molecule mu-opioid receptor (MOR) agonist developed by Hansoh Pharmaceutical for the treatment of postoperative pain. It is characterized as a G-protein biased agonist, designed to selectively activate the G-protein signaling pathway while minimizing the recruitment of beta-arrestin-2, a protein associated with opioid-induced side effects such as respiratory depression and gastrointestinal dysfunction. By favoring analgesic signaling over pathways that lead to adverse events, tegituridine aims to provide a safer alternative to traditional opioids like sufentanil. Clinical development, including Phase 3 trials, has focused on its efficacy in postoperative pain management and its impact on psychomotor recovery, particularly in vulnerable populations such as frail elderly patients undergoing laparoscopic surgery.
