Teglarinad is a water‑soluble small‑molecule prodrug of the nicotinamide phosphoribosyltransferase (NAMPT) inhibitor CHS‑828/GMX1778 that was developed for the treatment of solid tumors and lymphomas. By inhibiting NAMPT, the rate‑limiting enzyme in the NAD⁺ salvage pathway, its active metabolite depletes intracellular NAD⁺, disrupting energy metabolism, DNA repair, and survival signaling in rapidly proliferating cancer cells, leading to antitumor activity including interference with DNA repair and inhibition of angiogenesis.[1][3][9][12][7]
