Teglicarpidine fumarate (HS-10380) is a novel, small-molecule biased mu-opioid receptor (MOR) agonist developed by Jiangsu Hengrui Pharmaceuticals. It is designed to selectively activate the G-protein signaling pathway while minimizing the recruitment of β-arrestin-2, a mechanism intended to provide potent analgesia with a reduced incidence of typical opioid-related adverse effects such as respiratory depression and gastrointestinal dysfunction. The drug was approved by the NMPA in China in early 2024 for the management of pain. It is primarily administered via intravenous injection and is being further evaluated in post-marketing studies for procedural analgesia, including hysteroscopic surgery, often in combination with sedatives like remimazolam.
