Tegor, also known as Tegafur Gimeracil Oteracil Potassium, is an oral antineoplastic agent that is a fluoropyrimidine derivative. It is a combination drug comprising three active components: Tegafur (FT), Gimeracil (CDHP), and Oteracil (Oxo). Tegafur acts as a prodrug for 5-Fluorouracil (5-FU), which is a cytotoxic antimetabolite that interferes with DNA synthesis and RNA function. Gimeracil enhances the efficacy of 5-FU by inhibiting dihydropyrimidine dehydrogenase (DPD), an enzyme responsible for the degradation of 5-FU, thereby maintaining higher and more prolonged concentrations of 5-FU in the body. Oteracil reduces the gastrointestinal toxicity of 5-FU by inhibiting orotate phosphoribosyltransferase (OPRT) in the gastrointestinal tract, which decreases the phosphorylation of 5-FU in this region. This combination allows for sustained systemic exposure to 5-FU while mitigating its gastrointestinal side effects. Tegor is approved for the treatment of various cancers, including gastric cancer, colorectal cancer, non-small cell lung cancer, head and neck cancer, breast cancer, pancreatic cancer, and biliary tract cancer. It is administered orally.
