TEK-overexpressing lentivirus is a gene therapy construct designed to deliver and increase the expression of the TEK gene, which encodes the Tyrosine kinase with immunoglobulin and epidermal growth factor homology domains 2 (TIE2) receptor. TIE2 is a receptor tyrosine kinase primarily expressed on endothelial cells and some tumor cells, where it plays a pivotal role in angiogenesis, vascular stability, and survival. The lentiviral vector serves as a delivery vehicle to integrate the TEK sequence into the host cell genome, leading to sustained protein production. In the context of oncology research, such as cervical cancer, overexpressing TEK in tumor cells has been shown to promote tumor angiogenesis by upregulating TIE2 and VEGFR2 in the surrounding vascular endothelial cells via the TIE2-AKT/MAPK signaling axis. While primarily used as a research tool to study vascular biology and tumor progression, TIE2-targeted gene therapies are also explored for their potential in regenerative medicine to promote revascularization in ischemic tissues.
