Overview

**Tel-eVax** is a genetic cancer vaccine developed for veterinary use, primarily targeting canine tumors such as multicentric diffuse large B-cell lymphoma (DLBCL). It employs a prime-boost strategy using an Adenovirus 6 (Ad6) vector expressing a codon-optimized, catalytically inactive canine telomerase reverse transcriptase (dTERT, with D702A and V703I mutations), followed by DNA electro-gene transfer (DNA-EGT) of a plasmid encoding the same dTERT fused to a tissue plasminogen activator leader sequence and the B subunit of *E. coli* heat-labile enterotoxin (LTB) for enhanced immunogenicity. The vaccine induces strong anti-dTERT immune responses, including antibodies and cell-mediated immunity, without significant adverse effects. When combined with chemotherapy regimens like COP or CHOP, it significantly prolongs overall survival in dogs with DLBCL (e.g., 64.5 weeks with CHOP), outperforming historical chemotherapy-alone data, and shows promise for broader canine cancers and potential translation to human oncology targeting human TERT (hTERT).[1][3]
