Teleukin (teleukin; F16IL2) is an immunocytokine developed by Philogen S.p.A., consisting of a human single-chain antibody fragment (scFv) fused to human interleukin-2 (IL-2). The antibody portion, F16, specifically targets the extra-domain A1 of tenascin-C, a protein highly expressed in the extracellular matrix of the neovasculature of various solid tumors and in the bone marrow of patients with acute myeloid leukemia (AML). By binding to tenascin-C, Teleukin facilitates the localized delivery and concentration of IL-2 within the tumor microenvironment. This targeted delivery aims to stimulate the proliferation and activation of natural killer (NK) cells and T lymphocytes, thereby enhancing the anti-tumor immune response while potentially reducing the systemic toxicity associated with high-dose IL-2 therapy. In clinical development for AML, Teleukin has been investigated in combination with the anti-CD33 antibody BI 836858, particularly for patients who have relapsed after allogeneic hematopoietic stem cell transplantation (alloHSCT), to exploit the synergy between targeted cytokine stimulation and antibody-dependent cellular cytotoxicity (ADCC).
