Telisotuzumab adizutecan is an antibody-drug conjugate (ADC) developed for the treatment of advanced solid tumors. It consists of a c-Met–targeting monoclonal antibody (telisotuzumab) conjugated to a topoisomerase 1 inhibitor payload (adizutecan). The drug is designed to selectively deliver cytotoxic chemotherapy to tumor cells overexpressing the c-Met protein, which is implicated in tumor growth and progression. Telisotuzumab adizutecan has shown antitumor activity in multiple cancer types, including colorectal cancer, non-small cell lung cancer, and gastroesophageal adenocarcinoma. Its mechanism involves binding to c-Met on tumor cells, internalization of the ADC, and intracellular release of the topoisomerase inhibitor payload leading to DNA damage and cell death.
