Overview

Telisotuzumab vedotin is a first-in-class, c-Met protein-directed antibody-drug conjugate (ADC) developed for the treatment of non-small cell lung cancer (NSCLC). It consists of a humanized monoclonal antibody (ABT-700) targeting the c-Met receptor tyrosine kinase, which is often overexpressed in solid tumors. The antibody is conjugated to the cytotoxic agent monomethyl auristatin E (MMAE) via a valine-citrulline linker. Upon binding to c-Met on tumor cells, the ADC is internalized and MMAE is released intracellularly, inhibiting tubulin polymerization and inducing cell death. The drug has shown antitumor activity in NSCLC patients with c-Met overexpression, particularly those with EGFR wild-type disease.
