Overview

Telitacicept is a recombinant fusion protein that combines the extracellular domain of the transmembrane activator and calcium modulator and cyclophilin ligand interactor (TACI) receptor with the fragment crystallizable (Fc) region of human immunoglobulin G1. It is designed to target and neutralize two key B-cell activating cytokines—B lymphocyte stimulator (BLyS/BAFF) and a proliferation-inducing ligand (APRIL)—thereby suppressing the development and survival of plasma cells and mature B cells. This mechanism directly reduces pathogenic antibody production in autoimmune diseases. Telitacicept has been developed primarily for B cell-mediated autoimmune diseases such as systemic lupus erythematosus, rheumatoid arthritis, myasthenia gravis, multiple sclerosis, IgA nephropathy, neuromyelitis optica spectrum disorders, and Sjögren's syndrome. It was first approved in China for active systemic lupus erythematosus in March 2021; it has since also been approved there for rheumatoid arthritis and generalized myasthenia gravis[3][4][5][7].
