AD-209-1A is an oral fixed-dose combination (FDC) antihypertensive drug candidate developed by Addpharma. It contains 20 mg of telmisartan, an angiotensin II receptor blocker (ARB), and 2.5 mg of amlodipine, a dihydropyridine calcium channel blocker (CCB). Telmisartan lowers blood pressure by selectively antagonizing the angiotensin II type 1 (AT1) receptor, which prevents the vasoconstrictive and aldosterone-secreting effects of angiotensin II. Amlodipine inhibits the transmembrane influx of calcium ions into vascular smooth muscle and cardiac muscle, resulting in peripheral arterial vasodilation. AD-209-1A was utilized as an active comparator arm in a Phase II clinical trial evaluating the triple combination therapy AD-209 (telmisartan/amlodipine/chlorthalidone) for the treatment of essential hypertension in South Korea.
