TeloB-VAX is a novel cell-based therapeutic cancer vaccine designed to induce an immune response against cancer cells by targeting telomerase reverse transcriptase (TERT), an enzyme overexpressed in more than 85% of human cancers. The therapy uses the patient’s own circulating B lymphocytes, which are transfected ex vivo with a patented engineered plasmid DNA encoding telomerase antigens. After incubation, these modified cells are re-infused into the patient to stimulate a cytotoxic CD8+ T cell response specifically directed at tumor cells expressing telomerase. The approach aims to harness and direct the immune system to recognize and destroy both differentiated cancer cells and progenitor cancer stem cells. Initial development was by University of California San Diego; Adamis Pharmaceuticals (now DMK Pharmaceuticals) acquired rights and further developed the technology for oncology indications, particularly castrate-resistant prostate cancer. Early clinical studies have demonstrated safety, immunogenicity, and induction of tumor-specific cytotoxic responses[1][4][5].
