Telodipine is a third-generation dihydropyridine calcium channel blocker (CCB) developed by Shandong Xuanzhu Pharmaceutical Technology (a subsidiary of Sihuan Pharmaceutical) for the treatment of hypertension. Like other dihydropyridines, it acts by selectively inhibiting L-type calcium channels in vascular smooth muscle cells, preventing the influx of calcium ions and resulting in peripheral vasodilation and a subsequent decrease in blood pressure. Telodipine is designed to provide long-acting antihypertensive effects with a potentially improved safety profile compared to earlier CCBs. It has undergone clinical evaluation in China, reaching Phase 3 development for the management of essential hypertension, with a New Drug Application (NDA) accepted by the NMPA in early 2023.
