Drug intelligence / Profile preview
Telomeric antisense oligonucleotides (tASOs) are sequence-specific antisense oligonucleotides designed to target telomeric non-coding RNAs (tncRNAs), which are transcribed at dysfunctional or damaged telomeres. These tncRNAs are essential for recruiting DNA damage response (DDR) factors to damaged telomeres, thereby initiating and maintaining cellular senescence and chronic inflammation. By selectively binding to and silencing these tncRNAs, tASOs inhibit DDR activation at dysfunctional telomeres without altering telomere length. This therapeutic approach, originated by scientist Fabrizio d'Adda di Fagagna at IFOM and CNR, is being advanced clinically by spin-off TAG Therapeutics under the platform name TELOBLOCK. It has demonstrated preclinical efficacy in reversing cellular senescence, reducing inflammation, and restoring tissue homeostasis in models of idiopathic pulmonary fibrosis (IPF), Hutchinson-Gilford progeria syndrome (HGPS), aplastic anemia, and age-related hematopoietic decline.
