Drug intelligence / Profile preview
Temigal is a fixed-dose combination of sacubitril and valsartan developed by Provident Pharmaceuticals. It is a generic version of Entresto, which belongs to the class of angiotensin receptor-neprilysin inhibitors (ARNIs). Sacubitril is a prodrug that is metabolized in vivo to the active neprilysin inhibitor sacubitrilat (LBQ657), which prevents the degradation of natriuretic peptides (such as atrial natriuretic peptide [ANP] and brain natriuretic peptide [BNP]), thereby promoting vasodilation, natriuresis, and diuresis. Valsartan is an angiotensin II receptor blocker (ARB) that selectively blocks the angiotensin II type-1 (AT1) receptor, inhibiting the renin-angiotensin-aldosterone system (RAAS) to prevent vasoconstriction, aldosterone release, and cardiac remodeling. Temigal is formulated as film-coated tablets in strengths of 24+26 mg, 49+51 mg, and 97+103 mg, and is indicated for the treatment of chronic heart failure and essential hypertension.
