Overview

Temozolomide + β-elemene is an experimental combination therapy for malignant glioma and other brain tumors. Temozolomide is a well-established oral alkylating agent used in the treatment of glioblastoma and other high-grade gliomas. β-Elemene is a plant-derived small molecule extracted from Curcuma wenyujin, known for its ability to cross the blood-brain barrier, reverse multidrug resistance, and exert broad-spectrum antitumor effects. The combination has shown synergistic antitumor activity in preclinical models by inhibiting tumor growth more effectively than either agent alone. Mechanistically, temozolomide induces DNA damage leading to apoptosis in tumor cells, while β-elemene inhibits proliferation through modulation of signaling pathways such as ERK1/2-Bcl-2/survivin and enhances chemosensitivity to temozolomide[1][3]. This combination also alters pharmacokinetics by prolonging the mean residence time of temozolomide in brain tissue[1].
