Overview

TEDDI is an intensive immuno-chemotherapy regimen developed by the National Cancer Institute (NCI) for the treatment of primary central nervous system lymphoma (PCNSL). The regimen is a combination of five drugs: temozolomide (T), etoposide (E), pegylated liposomal doxorubicin (D, often referred to by the brand name Doxil), dexamethasone (D), and the Bruton's tyrosine kinase (BTK) inhibitor ibrutinib (I). This combination was specifically designed to address the activated B-cell (ABC) origin of most PCNSL cases and to utilize agents with favorable central nervous system penetration. Ibrutinib, a key component, targets the B-cell receptor signaling pathway which is frequently dysregulated in these lymphomas. The regimen is often evaluated in clinical trials in combination with rituximab, referred to as the TEDDI-R regimen.
