Overview

Temozolomide + osimertinib is an investigational combination therapy consisting of two oral small molecule drugs. Temozolomide is an alkylating agent that induces DNA damage, leading to tumor cell death, and is primarily used in the treatment of glioblastoma and other brain tumors. Osimertinib is a third-generation, irreversible epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor (TKI) that selectively targets both EGFR-sensitizing mutations and the T790M resistance mutation found in non-small cell lung cancer (NSCLC). The combination has been explored for its potential to treat central nervous system malignancies such as glioblastoma with EGFR alterations and NSCLC with leptomeningeal metastases, leveraging their complementary mechanisms—temozolomide’s cytotoxicity and osimertinib’s targeted inhibition of mutant EGFR signaling[1][2][3][4].
