A combination therapy consisting of temozolomide, an oral alkylating agent, and thalidomide, an oral immunomodulatory and antiangiogenic agent. Temozolomide damages DNA through alkylation, while thalidomide prevents new blood vessel formation and modulates immune responses. This dual-mechanism approach targets both cancer cells directly and their supporting vasculature, showing particular efficacy in neuroendocrine tumors, glioblastoma multiforme, and melanoma with brain metastases.
