Overview

This is a combination regimen of three drugs: temsirolimus, paclitaxel, and carboplatin. Temsirolimus is an mTOR inhibitor that binds to FKBP-12 and inhibits the mammalian target of rapamycin (mTOR), leading to cell cycle arrest in the G1 phase and inhibition of tumor cell proliferation[6]. Paclitaxel is a microtubule-stabilizing agent that disrupts mitosis by promoting tubulin polymerization. Carboplatin is a platinum-based chemotherapeutic that causes DNA crosslinking and apoptosis. This combination has been studied primarily in advanced solid tumors, including clear cell ovarian cancer, endometrial cancer, head and neck squamous cell carcinoma, and other malignancies suitable for platinum/taxane chemotherapy[1][2][3][5][7]. The rationale for combining these agents includes potential additive or synergistic effects due to their distinct mechanisms of action.
