The combination of temsirolimus and trebananib is an investigational drug combination that has been studied in clinical trials for advanced solid tumors. This combination targets both the ANG-Tie2 pathway (via trebananib) and the mTOR pathway (via temsirolimus). Trebananib (also known as AMG 386) is an inhibitor of ANG1/2-Tie2 interaction that may stop tumor growth by blocking blood flow to tumors. Temsirolimus is an mTOR inhibitor that may stop the growth of tumor cells by blocking certain cellular pathways involved in cancer cell proliferation. Combination therapy of temsirolimus and trebananib (AMG 386). Trebananib is an inhibitor of ANG1/2-Tie2 interaction that may inhibit tumor growth by blocking blood flow to tumors. Temsirolimus is an mTOR inhibitor that may stop the growth of tumor cells by blocking the PI3K/Akt/mTOR pathway. The combination was investigated based on evidence of crosstalk between the ANG-Tie2 and PI3K/Akt/mTOR pathways, with the hypothesis that combined blockade may have additive anti-cancer activity.
