Overview

Temtokibart is a fully human investigational monoclonal antibody that acts as an antagonist of the interleukin-22 receptor alpha-1 subunit (IL-22RA1). By blocking IL-22RA1, it inhibits signaling from the cytokine interleukin-22 (IL-22), which is implicated in skin barrier dysfunction and inflammation in atopic dermatitis. Temtokibart also partially inhibits IL-20 and IL-24 signaling. The drug represents a first-in-class approach for moderate-to-severe atopic dermatitis by targeting a different immunological pathway than existing biologics such as dupilumab (which targets IL-4/IL-13). It has demonstrated efficacy and safety in phase 2a and phase 2b clinical trials for adults with moderate-to-severe atopic dermatitis, showing significant improvements in Eczema Area and Severity Index (EASI) scores compared to placebo[1][2][3][5][6][7].
