Drug intelligence / Profile preview
ten-eleven translocation 3 small interfering RNA (siTET3) is a nanoparticle-coated or liposomal nanoparticle (LNP)-encapsulated small interfering RNA designed to target and silence ten-eleven translocation 3 (TET3), a DNA methylcytosine dioxygenase that acts as a DNA methylation eraser. In Group 3 medulloblastoma (G3-MB), TET3 is recruited by the transcription factor OTX2 to catalyze active DNA demethylation at the Otx2 super-enhancer, promoting chromatin opening, tumor cell proliferation, and stemness. By silencing TET3, the siRNA impairs this hypomethylation feedback loop, reducing OTX2 expression and suppressing tumor progression. Preclinical studies in patient-derived orthotopic xenograft models of G3-MB have demonstrated that LNP-delivered siTET3 effectively suppresses tumor growth.
