Tenapanor + omeprazole is a combination of two oral pharmaceutical drugs: tenapanor and omeprazole. There is no evidence of a fixed-dose combination product or any branded combination under this name; thus, this refers to the co-administration of two individual drugs. Tenapanor is a locally-acting small molecule inhibitor of the sodium/hydrogen exchanger isoform 3 (NHE3) on the apical surface of enterocytes in the intestine. It reduces absorption of sodium and phosphate, leading to increased intestinal sodium and water content, and is used for treatment of constipation-predominant irritable bowel syndrome (IBS-C) and to reduce serum phosphorus in adults with chronic kidney disease (CKD) on dialysis who are not adequately responding to or intolerant of phosphate binders[2][5]. Omeprazole is a proton pump inhibitor (PPI) that irreversibly inhibits the H+/K+ ATPase enzyme in gastric parietal cells, suppressing gastric acid secretion. It is used for the treatment of gastroesophageal reflux disease (GERD), peptic ulcer disease, and related indications. No unique mechanism of action or indication emerges from the simple co-administration of these two agents; any pharmacodynamic effects or interactions will reflect those of the individual components. Available evidence demonstrates that co-administration of omeprazole does not affect the safety or tolerability of tenapanor[1].
