Tenofovir amibufenamide is an investigational oral prodrug of tenofovir, designed as a nucleotide reverse transcriptase inhibitor (NRTI) for the treatment of chronic hepatitis B virus (HBV) infection. It is structurally and pharmacologically related to tenofovir alafenamide but features a different prodrug moiety intended to improve delivery and reduce systemic exposure. Like other tenofovir prodrugs, it is converted intracellularly to the active metabolite, tenofovir diphosphate, which inhibits HBV polymerase by causing DNA chain termination during viral replication. Tenofovir amibufenamide has been developed primarily in China and has undergone clinical trials for chronic hepatitis B.
