Drug intelligence / Profile preview
Tenofovir amibufenamide fumarate (TMF) is a novel, orally active nucleotide reverse transcriptase inhibitor (NRTI) and a second-generation prodrug of tenofovir. Developed by Hansoh Pharma, it was approved in China in June 2021 for the treatment of chronic hepatitis B virus (HBV) infection in adults. TMF is designed using ProTide technology, featuring an additional methyl group compared to tenofovir alafenamide (TAF), which enhances its plasma stability, lipid solubility, and cell membrane penetration. This optimization allows for efficient delivery of the active metabolite, tenofovir diphosphate, directly to hepatocytes, resulting in potent antiviral efficacy at a lower dose (25 mg daily) with reduced systemic exposure and improved bone and renal safety compared to tenofovir disoproxil fumarate (TDF).
