tenofovir disoproxil fumarate

Tenofovir disoproxil fumarate is an oral prodrug of tenofovir, a nucleotide analog reverse transcriptase inhibitor (NtRTI) used in the treatment of HIV-1 infection and chronic hepatitis B. After absorption, it is rapidly converted to its active form, tenofovir, which is then phosphorylated intracellularly to form tenofovir diphosphate. This active metabolite competitively inhibits viral reverse transcriptase and hepatitis B polymerase by mimicking deoxyadenosine 5'-triphosphate and causing premature DNA chain termination during viral replication. Tenofovir exhibits antiviral activity against retroviruses including HIV-1 and HIV-2 as well as hepadnaviruses such as HBV. It has a long intracellular half-life allowing for once-daily dosing. Tenofovir disoproxil fumarate was originally developed by Gilead Sciences and is marketed under the brand name Viread[1][3][4][5][6][8].
