Overview

Tenofovir exalidex is an orally active, liver-targeted, lipid-conjugated prodrug of tenofovir developed to treat chronic hepatitis B virus (HBV) infection. It is designed to enhance bioavailability and tissue penetration by leveraging lipid uptake mechanisms, resulting in higher concentrations in liver cells and lower systemic exposure compared to other tenofovir formulations. This targeted delivery aims to reduce the risk of renal and bone toxicities associated with traditional tenofovir therapies. Tenofovir exalidex acts as a nucleotide reverse transcriptase inhibitor, blocking viral replication by inhibiting the HBV reverse transcriptase enzyme. The drug has demonstrated potent antiviral activity against HBV in clinical studies and was also investigated for HIV infections but development for HIV has been suspended. Originally developed by Chimerix, it was further advanced by ContraVir Pharmaceuticals (now Hepion Pharmaceuticals)[3][4][6][7][8].
