Tenofovir gel is a topical formulation of the antiretroviral drug tenofovir, developed as a microbicide for the prevention of HIV infection. It is most commonly studied as a 1% vaginal or rectal gel. Tenofovir is a nucleotide reverse transcriptase inhibitor (NRTI) that acts by inhibiting viral reverse transcriptase, thereby blocking HIV replication. The CAPRISA 004 trial demonstrated that use of the vaginal gel reduced HIV acquisition in women by approximately 39%. Tenofovir gel has also been evaluated for rectal use in men and women at risk for HIV infection. Despite promising results in early trials, development was discontinued after Phase 3 studies due to insufficient efficacy and adherence challenges[1][2][4][7].
