Tepinoroth is a novel topical hydrocarbon receptor (HR) agonist, likely targeting the aryl hydrocarbon receptor (AhR), currently under investigation for the treatment of psoriasis. It exerts durable immunomodulatory effects by modulating mitochondrial activity in immune cells, which leads to metabolic quiescence and a reduction in the production of mitochondrial reactive oxygen species (ROS). Furthermore, it decreases damage-associated molecular pattern (DAMP) signaling, thereby suppressing the inflammatory pathways associated with psoriatic lesions. The agent has been evaluated in human skin graft models to assess its efficacy in restoring skin barrier function and reducing cytokine-mediated inflammation.
