Overview

Tepoditamab (MCLA-117) is a bispecific monoclonal antibody designed to target both CLEC12A, a myeloid inhibitory C-type lectin-like receptor expressed on acute myeloid leukemia (AML) blasts and leukemic stem cells, and CD3 on T cells. By binding to these two targets, tepoditamab redirects and activates T cells to induce cytotoxic lysis of CLEC12A-positive AML cells. The antibody features a silenced Fc region to minimize non-specific activation of immune effector functions. Preclinical studies have shown that MCLA-117 induces T cell activation, proliferation, cytokine release, and robust killing of AML blasts while sparing normal hematopoietic progenitors. It is being developed primarily for the treatment of acute myeloid leukemia[1][4][5][6][8].
