Overview

Teprenone (also known as geranylgeranylacetone) is a small molecule drug used primarily for the treatment of gastric ulcers and gastritis. It acts as a cytoprotective agent by enhancing the integrity of the gastric mucosa through several mechanisms. Teprenone stimulates the synthesis of protective factors such as prostaglandins and mucus, increases bicarbonate secretion, promotes blood flow in the gastric mucosa, and induces heat shock protein 70 (HSP70), which helps protect cells from stress and injury. Unlike acid-suppressing drugs, it does not significantly affect normal physiological functions or acid secretion in the stomach. Teprenone is approved for use in several Asian countries (notably Japan) for treating peptic ulcer disease and gastritis[1][2][4][5][6]. **Developers:** Eisai **Manufacturers:** Eisai, Sawai Pharmaceutical Teprenone is especially notable for its use as a gastroprotective agent to prevent NSAID-induced gastrointestinal injury[5]. It has been widely studied in Asia but is not approved or commonly used outside this region.
