Drug intelligence / Profile preview
terbinafine + fluconazole is an investigational oral antifungal combination regimen being evaluated for the treatment of refractory dermatophytosis (fungal skin infections such as tinea corporis and tinea cruris). The combination consists of two small-molecule antifungal agents with complementary mechanisms of action targeting the fungal ergosterol biosynthesis pathway. Terbinafine is an allylamine antifungal that inhibits squalene epoxidase, preventing the conversion of squalene to lanosterol, while fluconazole is a triazole antifungal that inhibits lanosterol 14-alpha-demethylase (CYP51). By targeting different steps of the same biosynthetic pathway, the combination aims to achieve synergistic fungicidal activity and overcome treatment resistance. The regimen is being evaluated in clinical trials sponsored by Chittagong Medical College.
