Tergenpumatucel-L is an allogeneic whole-cell cancer immunotherapy developed by NewLink Genetics (now Lumos Pharma) for the treatment of non-small cell lung cancer (NSCLC). It consists of three irradiated, genetically modified human NSCLC cell lines (HAL-1, HAL-2, and HAL-3) that have been engineered to express the mouse gene for alpha(1,3)-galactosyltransferase. This enzyme catalyzes the addition of alpha-galactosyl (αGal) carbohydrate moieties to the cell surface proteins and lipids. Since humans naturally possess high titers of anti-αGal antibodies, the injection of these cells triggers a hyperacute rejection response. This innate immune activation is intended to recruit and activate antigen-presenting cells, which then process and present a broad spectrum of lung cancer-associated antigens to the patient's T cells, thereby stimulating a systemic, adaptive anti-tumor immune response against the patient's own cancer cells.
